CY5 and sulfonated CY5 in click reactions

Click reaction is a specific chemical reaction by which both CY5 and sulfonated CY5 can be efficiently attached to other molecules. Sulfonated CY5 has a wider application in click reactions in organisms due to its excellent properties in aqueous solution.

 

CY5 and sulfonated CY5 can be used in click reactions to label molecules containing alkyne functional groups to achieve their click reactions with doses containing azobenzene functional groups to form stable covalent linkages.

 

The steps for clicking on a response are as follows:

 

1. Marker preparation: Covalent binding of molecules containing alkyne functional groups to CY5 or sulfonated CY5. This can be accomplished by a chemical reaction (e.g., NHS ester reaction) or an enzyme-catalyzed coupling reaction. The introduction of CY5 or sulfonated CY5 into the molecule results in a fluorescent label.

 

2. Click reaction: Reaction of a dose containing an azobenzene functional group with a labeled molecule. The click reaction is usually copper-catalyzed and forms a stable triazacyclic linkage by reacting the alkynyl groups of the two functional groups with the azobenzene functional group.

 

3. Visualization: After the click reaction is complete, the fluorescent signal of the labeled material can be observed by viewing the sample using a fluorescence microscope or other fluorescence imaging instrument.The fluorescence of CY5 and sulfonated CY5 emits a reddish color, so that the labeled material can be localized and visualized by the red fluorescence.

 

Click reaction-conjugated CY5 or sulfonated CY5 fluorescent markers can be used to label biomolecules such as proteins, peptides, and nucleic acids for their localization, tracking, and visualization. This method is widely used in biology and biomedical research, such as protein interaction studies, cellular tracing, disease labeling, etc. The red fluorescent properties of CY5 and sulfonated CY5 make them ideal for studying biomolecules.
