Cyanine dyes (Cy) are a commonly used class of near-infrared fluorescent dyes, and the spontaneous fluorescence of cells and tissues is small in the near-infrared band, providing higher specificity and sensitivity. The Cy series of fluorescent dyes are divided into non sulfonic cyanine dyes and sulfonic cyanine dyes. Their spectral properties are basically the same, and their reactions with protein small molecule modifications are also basically the same. The difference lies in their solubility. Non sulfonated cyanine dyes must be dissolved in organic solvents such as DMF or DMSO for use, and buffered aqueous solutions must be added for use. The sulfonic cyanine dye is dissolved in water and the entire use process is carried out in the aqueous phase. Cy5.5 is a commonly used fluorescent dye that can be used for cell staining as well as in vivo imaging of small animals. Dyes with similar fluorescence characteristics include Alexa Fluor 680, Dylight 680, etc., which excite X: 680nm and emit 698m light coefficient (emax): 209000
